Rose Bengal
Plate Antigen (RBPT)

Description : The
antigen is a suspension of pure
smooth culture of Rose Bengal dye
stained Brucella abortus strain 99
in phenol saline. The antigen is
used for the plate test for
serological diagnosis of Brucella
infection. |

Kit : Contains the antigen, positive
serum and a negative serum
Testing
: Collect the blood and separate the
serum. One drop of serum is put on a
glass slide. One drop of antigen is
then added to the serum. The serum
and antigen are then mixed quickly
using a tooth pick or a match stick
(use separate tooth pick for each
serum sample). The slide is held
over a white surface and rocked
gently from side to side for 4 to 5
minutes. A strongly positive
reaction will be visible during
first 30 seconds as distinct clumps.
Storage
:
The antigen should be
stored at 4oc.expiry date is 9
months from the date of manufacture

Brucella
Milk Ring Test Antigen (MRT)
Description : The antigen is a
suspension of pure smooth culture of
stained Brucella abortus strain 99
stained with hematoxylin.
Use
: The
antigen is used to test the presence
of Brucellosis in a herd, in
individual and in antibodies in
milk. The test can be made on pooled
cow milk from a herd, milk from
tankers, composite sample or quarter
milk sample from individual cow and
/or milk sample collected from
individual sheep and goat. The
usefulness of this antigen in
testing the buffalo milk is not well
defined.
Storage:
Antigen should be stored at 4oc
. The expiry of potency is 9 months
from the date of manufacture.
Test :
Well mix the milk
sample are taken in a narrow (11mm X
100mm) test tubes. One drop(0.03ml)
of the antigen is added. The
contents are mixed gently by closing
the several time. The mixture is
allowed to stand for 1 minute to
make sure that the antigen is
thoroughly mixed with the milk. The
tubes are incubated at 37oc for 1
hour.
The test is read
using an uniform light source. If
the blue colour in the cream layer
at the top of the fluid column is
deeper than the remaining milk
column (ie , presence of a coloured
ring) the test is considered
positive.
If the intensity of
colour in the cream layer is equal
to or less than in the milk portion,
the test is considered negative.

Salmonella Pullorum Coloured
Antigen

Description :
The antigen is
a suspension of pure smooth culture
of Crystal violet dye stained
Salmonella pullorum strain
17 in a solution containing 1%
formaline, 1% Potassium dihydrogen
phosphate and 0.85% Sodium Chloride |

Uses :
The antigen is used for the
detection of pullorum infection in
birds.
Test
procedure :
About one ml of blood is drawn from
the wing of each birds. Equal amount
of freshly drawn blood and antigen
are mixed on a glass plate by means
of a match stick. The plate is
tilted back and forth. the presence
of clumps is detected by the help of
light or white paper in the
background. The agglutination should
appear within 1 minute, if the blood
is from fowl positive for salmonellosis.
Storage :
It should be stored at 4oc. do not
freeze the antigen. expiry date is 6
month from the date of manufacture.
